miR-137 prevents inflammatory response, oxidative stress, neuronal injury and cognitive impairment via blockade of Src-mediated MAPK signaling pathway in ischemic stroke

class="figure-viewer-img"

Figure 3. Inhibition of p38 or ERK2 signaling pathway attenuates oxidative stress and the inflammatory response in the brain of MCAO mice. (A) detection of ROS in brain tissues from mice treated with si-p38, si-Erk2 or in combination using DCFH-DA fluorescent probe; (B) detection of TNF-α, IL-1, IL-6, IL-17 and IL-10 protein expression in the brain tissues of mice treated with si-p38, si-Erk2 or in combination by ELISA; (C) correlation analysis between the score of neurological function and contents of TNF-α, IL-1, IL-6, IL-17 and IL-10 in brain tissues of mice treated with si-p38, si-Erk2 or in combination. Measurement data are expressed as mean ± standard deviation, which were compared by one-way ANOVA, followed by Tukey's post hoc test. Correlation analysis was performed using Pearson correlation coefficient, r > 0 represents a positive correlation, r < 0 represents a negative correlation, p < 0.05 was indicative of statistical significance; * p < 0.05 vs. control group (sham-operated wild-type mice); # p < 0.05 vs. MCAO group (wild-type mouse models of MCAO). N = 15.
